Cholesterol oxidation products, known as oxysterols, are potentially involved in the pathogenesis of major chronic diseases, including atherosclerosis, Alzheimer’s disease, and intestinal disease (including colon cancer). Oxysterols are involved in various key steps of these complex processes, mainly thanks to their ability to act through up-regulation of oxidative stress, inflammation, and cell toxicity, however, their role needs still to be further investigated.
This project aim to better investigate the oxidative, inflammatory, and cytotoxic effects of these products on vascular, neuronal or intestinal cells through the modulation of various cellular pathways and/or key molecules. Their involvement in the pathogenesis of these diseases will be confirmed using specific antioxidant or antiinflammatory molecules as well as gene silencing. In vitro, ex-vivo, and, eventually, in vivo experiments will be performed; all the involved molecules will be analyzed by molecular biology, immunohistochemistry, and microscope techniques.
A better understand of all the molecular mechanisms and molecules involved in the pathogenesis of these inflammatory diseases might help to develop new drugs and/or therapeutic strategies.

The General Pathology Unit, is located in the Biological Center of the Department of Clinical and Biological Sciences, San Luigi Gonzaga Hospital, Regione Gonzole 10, Orbassano.
The laboratories are equipped for the updated and complete development of cell cultures, biochemical, biomolecular and cytohistological analyzes. The main equipment: quantitative RT-PCR, confocal microscope, multiplex technology.
