We have described previously the genomic activity of ERα in breast cancer and discovered that ERα controls transcription of protein-coding, as well as several long-noncoding RNA (lncRNAs) genes also in the absence of estrogen. Importantly, these genes control the luminal epithelial phenotype of these cells.
We are currently moving in two directions. From one side, we are expanding the model systems of Breast Cancer and working to understand how some important lncRNAs act and interact with other known molecular entities in order to control cell proliferation and differentiation. On the other side, we are moving to different model systems, e.g. lymphocytes, macrophages, adipocytes, neurons, to verify whether unliganded actions of ERα are important also in these cells in anaestrogenic conditions such as, for example, in post-menopausal women, or in women treated with aromatase inhbitors.
Our studies include epigenomics (i.e. ChIP-seq analysis of genome-wide Transcription Factor binding or Histone PTMs, ATAC-seq) as well as transcriptomics (long- and short- RNASeq, GRO-seq). Moreover, we have a very strong drive to bioinformatics and network analysis, that we run in collaboration with the Computer Science Dept.
