Spinocerebellar ataxias (SCAs) include more than 40 different types of diseases of the central nervous system. The incidence of SCAs is 1 every 30,000 subects and at present there is no cure. Our laboratory is studying animal models of SCA27 and SCA38. The latter is caused by mutations of the gene ELOVL5. The aim of our study is to understand the pathogenic mechanism and its connections to new physiological roles of Elovl5. The experimental techniques used for this study are: i) motor tests to follow up the evolution of the disease in the animal models; ii) immunohistochemistry in brain sections of the animal models; iii) electrophysiological recordings of membrane potential and ionic currents by the patch-clamp tecnique in brain slices from animal models; gene expression by quantitative RT-PCR. Our studies are aimed at aquiring knowledge necessary to design new therapeutic approaches. A second line of research regards the role of cerebellum in autism, studied in animal models with genetic modifications frequently found in patients. Finally, we study the mechanisms underlying the control of anxiety and depression by a novel subunit of voltage-dependent sodium channels.
